Anti-pagoclone

I have spoken extensively about Pagoclone, a drug that is currently undergoing trials for stuttering. One important issue is whether Pagoclone increases fluency, because anxiety is reduced or because somehow more control of the speech system exists. I believe that anxiety-reducing effects are a significant part of the effect. Not so much for low-stress situations, but for stressful situation.

I am wondering what happens if people get an anti-Pagoclone, e.g. if it reduces the level of X, then a drug that increases the level of X? If anti-Pagoclone does not cause stuttering in normal people, I would guess that anxiety-reducing effects are most important? Or not?

I am curious about what others taking pagoclone have found. I am in my third month of it and have noticed little difference and zero side effects? What do other people say?

I have been on Pagoclone for a couple of months, and have noticed improvement that began within the first few days and has continued. Family and Colleagues have commented that I am significantly more fluent.
Before, I was having difficulty and was uncomfortable is most speaking situations. Now I am comfortable and probably 95% fluent in dinner conversations, work sessions, and the normal speaking situations that come up in everyday life.
When I do have a block, it seems to be on certain words like my name. These blocks are easier than they were before.
As far as side effects, I think I am noticing a general sense of calm that was not there before.

I am currently six weeks in the trial, phase II. I have noticed much less blocking in my speech. I have noticed that my stuttering has not gone away but it is much less frequent. While I am in a block, the stress involved is much less. This is probably due to the anti-anxiety properties of Pagoclone. The side effects are minimal.

This is all exciting information for the rest of us waiting on the results of pagoclone. I was supposed to be part of the study but when I showed up the day of my appointment, the office said that the study closed the day before. Can't wait to hear more information this summer from Indevus.

I've been on it for a little over three months now and have noticed significant improvement in my speech. I used to get blocked on the phone with my name, but am able to 'plow' through much more quickly. I still have espisodes, but they're much smaller. I find myself relaxing more and breathing more before I get ready to talk. I am remembering to do things like relax and drop my shoulders. My confidence is also growing, which is fantastic.

All this information if very encouraging. I have for years thought that there would never be a cure and that I needed to live with this demon. But I see that will not be the case. Are there any other trials coming? does anyone knows? gabbiano7070@hotmail.com

My son age 16 has tried everything including the Speech Easy ear device to control his stuttering. Being in High School he is stressed alot and the stuttering makes it wors-interfereing in his social life and confidence. I am hoping that when this drug comes available he would be of age to try it. What are the long term side effects? He is quite hormonal at this age-so maybe the calming effect will help his up and down personality. I'm sure he has a chip on his shoulder due to his stuttering since his younger brother and sister are quite fluent. I feel he resents them, and therefore treats them badly. We need help to conquer this problem for him and get him to start living and enjoying his life. You're only young once and this hasn't been an enjoyable experience for him.

Thanks a lot to everyone for sharing their experience with pagoclone. Can anyone help me - where I can purchase pagoclone in South Africa. Or where one can buy it in USA - my friend is in USA at the moment and I can ask him.

Thanks

I can't believe there is finally a possible cure or help for this kind of stuttering. I am 56 years old and the hardest thing for me to say was my name when asked.I thought I was the only one with this problem. When I was younger my father wouldn't accept the fact that if I could talk good some times I should be able to talk good all the time. What a relife to know there are other people out there with the same problem. I would love to see this drug available. Any help in getting this would be most greatful. I am much better than I used to be but the PHONE was the biggesst panic attack of all.

For everyone inquiring about how to get Pagoclone - you can't. It's currently in clinical trials and, if all goes well, should be available to the public in ~2010.

Hi,

Very informative thread, thanks to all posters. Has anyone used olanzapine (Zyprexa) to treat stammering? I have heard it mentioned before on other websites, any more info from here would be great

wow finally the miracle drug for us stutterers. cant wait til its available the trials look really promising.

As for olanzapine, I would not recommend this. I tried it once and blacked out and saw rainbow stars like a normal stand up too fast thing, but much more intense and lasted around 10mins.

-very very scary and i would never try again

I have tried quetiapine (seroquel) which is a similar but less strong drug. its effects were not unpleasant like zyprexa but only mildly helpful with my stutter, similar to any sedative

I am on week 2 of the drug trial, the first week I noticed mild headache and being somewhat tired, but that has gone away. I've noticed an expanded awareness so far, kinda like thoughts flow easier and speaking feels easier. It's still early days, I'll keep you updated.

Is ths drug available, or is it still in trial?

Kaboul said...

Yes the drug is still in trial. I am currently in the final 2 months of the current trials, and there is a possibility that they will make another trial period before they bring pagoclone out on the market or not. That all depends on the FDA. I will begin the open label phase in the month of December and will be given the real medication until it comes out on the market (there is a big chance that I am on placebo right now as I haven`t noticed any changes in my speech and no side effects at all).

I will keep you guys updated on the results I am getting on the real meds and I will let you know if I have any information if there will be another trial period so you guys can sign up. From what I`ve heard from others in the trials, the results look really promising and some call it even life changing, so I doubt that the meds will be discontinued, and I expect them to come out on the market hopefully during 2010-2011.

October 22 2009 seems to be the last comment posted regarding pagoclone and it's clinical trial. Does anyone know the final results of the trial and if its still ongoing or not? I am 34 years old and have stuttered my entire life. If anyone can guide me to the location of this drug, I would be very grateful. etshaw25@yahoo.com

Hello to everyone,

I haven't posted here in a while. I am no longer in the pagoclone study, and I believe the study is over. I stayed throughout all of it, but decided to stop using the medication when I received a call to tell me that the trials will be stopped due to poor results. As for me, I didn't think that the medication helped me at all, and maybe the dose that they were giving us was too low, but I guess we'll never know. Hopefully something else is brewing out there that will help stutterers in the near future.

Best regards, stay positive!
